Predicting absorption of fragrance Chemicals through human skin

چکیده
In the endeavor to identify a mathematical model predictive of the mostly unexplored skin penetration of fragrance chemicals, the Potts-Guy equation was tested here by correlating the published, measured Kp for 20 such compounds with predicted Kp values. The data, covering a broad range of structures, are well described by that model (r 2 = 0.86, p < 0.001). Also, calculated octanol-water partition coefficients, an alternative in case of missing measured values, appear well founded when analyzed statistically: comparison of 33 measured partition values with the calculated log Ps show near-perfect correlation for the intermediate lipophilicity range typical of fragrance hemicals (0.5 < log P • 3.5; r 2 = 0.97, p • 0.001). The Potts-Guy relation thereby recommends itself as a simple and reliable method for the quantitative evaluation of human skin absorption potential of small-molecular-weight nonelectrolytes, and of fragrance chemicals, in particular where such data are largely missing.

Fragrance materials are widely used in cosmetics and other consumer products. The Research Institute for Fragrance Materials (RIFM) evaluates the safety of these ingredients and skin absorption is an important parameter in refining systemic exposure. Currently, RIFM's safety assessment process assumes 100% skin absorption when experimental data are lacking. This 100% absorption default is not s...

The article "Scents & Sensitivity" [EHP 106:A594-A599 (1998)] provides a good summary of the issues involved in concerns and controversies over fragranced products. However, several points should be addressed. The health effects of fragrances are a general health issue, an indoor air quality issue, an access issue, and an environmental issue. Unfortunately, the only issue the fragrance industry...
متن کاملStereoselective Permeation of Tretinoin and Isotretinoin through Enhancer-Treated Rat Skin. I. Effect of Ethanol and Sodium Dodecyl Sulfate
Many properties of chemicals depend on their streochemistry. Among these, the effects of streoisomerism on percutaneous absorption of drugs, which is subject of the present investigation, is not well studied yet. In this study, tretinoin (TT) and isotretinoin (ITT) (geometric isomers) were chosen and their permeations (alone or in the presence of each other) through enhancer-treated excised ...

متن کاملMetabolism of fluroxypyr, fluroxypyr methyl ester, and the herbicide fluroxypyr methylheptyl ester. I: during percutaneous absorption through fresh rat and human skin in vitro.
Percutaneous absorption of pesticides is a major determinant for risk assessment. Furthermore, cutaneous metabolism plays a role in penetration of certain chemicals. Therefore, the aim of these studies was to determine the transdermal metabolism of three related compounds [the herbicide, fluroxypyr methylheptyl ester (FPMH), fluroxypyr methyl ester (FPM), and fluroxypyr (FP)] during penetration...
